Les mollusques congelés représentent une part de plus en plus importante de nos importations de produits de la mer. Destinées à la consommation directe ou à la transformation, les noix de coquilles Saint Jacques congelées sont importées du Japon, du Royaume Uni ou d'Australie, en général glazurées individuellement, et souvent après trempage préalable destiné à les faire absorber une certaine quantité d'eau. C'est ainsi que l'eau totale ajoutée à la noix (glaçage + trempage) peut représenter 50 % du poids du produit avant traitement. Characterization, chemical modifications and in vitro anticoagulant properties of an exopolysaccharide produced by Alteromonas infernus ArchiMer

A new low-molecular-weight 'heparin-like' component was obtained from an exopolysaccharide produced by a mesophilic strain found in deep-sea hydrothermal vents. Data concerning the structure of the native high-molecular-weight exopolysaccharide (10(6) g/mol, 10% sulfate content) are reported for the first time. Two depolymerization processes were used to obtain low-molecular-weight (24-35 x 10(3) g/mol) oversulfated fractions (sulfate content 20 or 40%), Nuclear magnetic resonance studies indicated that after sulfation (40%), the low-molecular-weight fraction obtained by free radical depolymerization was less sulfated in the 6-O-position than the fraction depolymerized by acid hydrolysis. Polysaccharide labelling: impact on structural and biological properties ArchiMer

Three sulphated polysaccharides labelled at the reducing end with three different chemical treatments (2-aminopyridine (2-AP), 8-amino-1,3,6-trisulphonic acid (ANTS) and 6-(biotinyl)-aminocaproyl-hydrazide (BACH)) were compared. Molecular weight, chemical composition, nuclear magnetic resonance features and anticoagulant activity expressed as activated partial thromboplastin time were studied to determine whether structural and biological properties were retained or not after labelling. Effect of an oversulfated exopolysaccharide on angiogenesis induced by fibroblast growth factor-2 or vascular endothelial growth factor in vitro ArchiMer

The aim of this study was to determine the angiogenic properties of an oversulfated exopolysaccharide (OS-EPS) derived from a polysaccharide secreted by the mesophilic bacterium Alteromonas infernus. We compared the effect of this OS-EPS with that of a non-oversulfated exopolysaccharide (EPS) on human umbilical vein endothelial cell (HUVEC) proliferation, migration and differentiation induced by basic fibroblast growth factor (FGF-2) or vascular endothelial growth factor (VEGF). To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The growth and differentiation of bone cells is controlled by various factors which can be modulated by heparan sulphates. Here, we investigated the effects of an oversulphated exopolysaccharide (OS-EPS) on bone. We compared the effect of this compound with that of a native exopolysaccharide (EPS). Long-term administration of OS-EPS causes cancellous bone loss in mice due, in part, to an increase in the number of osteoclasts lining the trabecular bone surface. Some marine bacteria collected around deep-sea hydrothermal vents are able to produce, in laboratory conditions, complex and innovative exopolysaccharides. In a previous study, the mesophilic strain Alteromonas macleodii subsp. fijiensis biovar deepsane was collected on the East Pacific Rise at 2600m depth. A previously reported bacterial bioemulsifier, here termed microbactan, was further analyzed to characterize its lipid component, molecular weight, ionic character and toxicity, along with its bioemulsifying potential for hydrophobic substrates at a range of temperatures, salinities and pH values. Analyses showed that microbactan is a high molecular weight (700 kDa), non-ionic molecule. Gas chromatography of the lipid fraction revealed the presence of palmitic, stearic, and oleic acids; thus microbactan may be considered a glycolipoprotein. Genome sequence of Vibrio diabolicus and identification of the exopolysaccharide HE800 biosynthesis locus ArchiMer
